Writer isn’t an occupation, it’s a medical condition. You just put it in the wrong box.

What I was trying to say is that it is too long to paste here. I can paste the beginning and end:

I tried to post it here

I wish, I don't even work with fish (yet!)

Does your lab have those rims to prevent things from falling during an earthquake?

I never saw it on the east coast, but if you have them I have some pipette holders and kimwipe holders that hang off of them. The “hook” is generalizable to hang anything though (i hung a computer monitor too).

90-95% of what I print “holds” something. Pipettes, fish brains, name tags. Its funny, but irreplaceable.

My Printer (Eufymake/Ankermake) is nice, but i think they stopped making them. That being said, Black Friday is the best time to buy. I got mine refurbished for $116, a $500 printer. Two years later, still runs like when I got it.

Very nice paper in general. I think I will play with the growth phase. Perhaps pick tiny colonies and harvest early (from a large culture).

Wow, maybe I am lucky it inserted in the middle of my construct!

They are totally different plasmids. Ill check out the references, TY!

Horribleness, it is NOT okay!
November 19, 2025 at 3:28 PM
@addgene.bsky.social have you found some ways to deal with this?
November 19, 2025 at 3:24 PM
I found the TE consensus target sequence is quite common. So I suspect it isn’t as advertised. There is probably more preference for certain types of sequences. Possibly combined with growth conditions or something. IDK

That’s what I was thinking, but which one. Cloning for 15 years I only encountered this a few months back. It was trying to midiprep a plasmid from another lab. They tried many strains and still had the issue. Now it is happening while I am trying to clone… 😢

I used NEBuilder. But it is a transposon insertion from the e coli genome! It happens after assembly in the cells. So how to stop it?!
November 19, 2025 at 2:48 PM
It is not my insert. I had this happen with another plasmid. It is a transposon insertion from the e coli genome! So I have no idea how to prevent it, or minimize the probability.

how do they find out?

Maybe this is being added after you read the review and figured out who the reviewer is.
(Its better at the beginning, but adding it at the end leaves your soup with more of the volatile chemicals) Maybe both is best!
