Innate immune and metabolic signals induce mitochondria-dependent membrane lysis via mitoxyperiosis
Wang et al. identify a new lytic cell death pathway, mitoxyperilysis, triggered by the synergy of innate immune activation and metabolic stress. This process is regulated by mTORC2 and involves persistent mitochondria-membrane contact, leading to localized oxidative damage and cell lysis, with implications for inflammatory diseases and cancer therapy.

And the saga continues! Our collaborative work with outstanding Justin Korfhage & Thomas Melia’s lab in PNAS shows that ATG2A transfers triglycerides and does so bidirectionally! An exciting twist in our understanding of lipid transport.

ATG2A–DGAT2 cooperation fuels lipid droplet growth
Nature Structural & Molecular Biology, Published online: 27 November 2025; doi:10.1038/s41594-025-01720-4Cells store excess fat in lipid droplets to avoid lipotoxicity and maintain homeostasis. We identified an autophagy-independent role for the autophagy lipid transfer protein ATG2A in helping direct lipids to growing lipid droplets and promoting recruitment of the enzyme DGAT2. This coordination enhances triglyceride storage, protects the endoplasmic reticulum from lipid overload and limits the misrouting of lipids into other metabolic pathways.
