The ground truth for AD is a bit of a debate recently, to say the least! But I am a big fan of the intersection between a clinical diagnosis and a biological definition.

- we need to learn from system biology regarding the interpretation of gene hits. For too long we have relied on imprecise tools (gene ontology) that are often wrongly annotated and cannot sufficiently account for the redundancy of biological systems or the existence of feedback loops

Interesting read! Some thoughts: - a GWAS is only as good as the patient inclusion. There has been immense efforts for AD on that aspect, even doing small postmortem GWAS on path confirmed cases
- the absence of a coherent picture at the gene level might indicate multiple disease mechanisms
