A meta-analysis confirms that homeostatic and immunogenic mature signatures are conserved across tissues and DC subsets, supporting a unified nomenclature for mature DCs.

Our study revealed that empty LNPs do not induce immunogenic maturation of DCs, while mRNA-containing lipids do, showing that the cargo, not the lipids, determine adjuvanticity.

By using an approach relying on the induction of homeostatic DC maturation by uptake of empty LNPs and immunogenic DC maturation by TLR ligand-adjuvanted LNPs, and performing CITE-SEQ, a flow cytometry panel was designed to assign mature DCs as immunogenic or homeostatic.
