Collagen secretion, membrane trafficking

Formerly @crg.eu
Nice paper from Kota @saitolab.bsky.social! Balanced phosphorylation of TANGO1 and Sec16A keep ER exit sites stable and functional.
Do cells maintain this dynamically to keep secretory machinery primed and responsive to changing cargo loads or environmental signals?

#MembraneTraffickers! Our next online seminar is this Thursday, Nov 6 at 5 PM CEST!
We’ll hear from @agatawitkowska.bsky.social on mechanical control of neurotransmission and @abdourachidthiam.bsky.social on lipid storage mechanisms in health and disease.
Spread the word and join the discussion!

How do resident Golgi proteins recycle during cisternal maturation? Nice preprint from Adam Krahn in the Glick lab shows different classes of intra-Golgi vesicles in yeast: two COPI-dependent (early and mid) and one late clathrin/AP-1/Ent5 pathway.

Interesting preprint from Ginny Farias! Axons locally synthesize transmembrane proteins that traffic via axonal ERES, bypassing the Golgi, using HDLBP and NRZ tether-SEC22B. This unconventional secretion is crucial for axon growth and bouton assembly.

Another interesting preprint on the mechanobiology of Golgi export! Matrix stiffness boosts secretion via a Src-FAK-AMPK-GBF1 pathway, controlling Golgi cargo sorting and preventing lysosomal degradation, linking mechanics to trafficking.

Exciting preprint on organelle mechanoadaptation from Stephanie Miserey and Felix Campelo @felixmendu.bsky.social! Extracellular forces modulate Golgi secretory output by tuning carrier biogenesis via microtubule acetylation, DAG, and PKD. Golgi is a mechanoregulator!

Gutted to hear of Prof PP Divakaran’s passing. Friend, mentor, drinking companion; a trove of insights on physics, ancient math, politics, architecture. He loved food, wine and conversation; each talk full of curiosity and humour. Endlessly generous with mind and company. He will be deeply missed.

Nice preprint from Basto lab (Curie)! Polyploid Drosophila salivary gland cells eliminate centrosomes via novel MyoII- and autophagy-dependent exocytosis: preserves mitochondrial function and supports high metabolic demands. Interesting secretory autophagy pathway!

Golgis of the world, unite! You have nothing to lose but your ribbons. If it can shape the secretory pathway, it can surely earn a place in the dictionary.

Glad to be part of this study from Meir Aridor! COPII assembles at ER exit sites when the ERGIC/Golgi is close by!

p125A links COPII outer coat assembly to PI4P-rich Golgi membranes, coordinating tunnel-based collagen secretion from the ER to the ERGIC/cis-Golgi.

Interesting preprint from Liang Ge! TMED proteins translocate unconventional (cytosolic) secretory cargoes into the ERGIC with HSP90 aid. ERGIC localisation and cargo-binding drive TMED homo-oligomerization enhancing translocation, hetero-tetramerisation inhibits it.

Nice C. elegans preprint from Melissa Cizeron (Bessereau group, Lyon)! CLE-1 (col XV/XVIII orthologue) is a scaffold, anchoring synaptic Punctin. The complex ensures proper receptor placement and synapse identity. Loss of CLE-1 scrambles receptor localization.

Bienvenue à Paris to our newest lab member, Dr Samruddhi Shembekar!
After a PhD on how mitochondria stay intact while moving through the cell, featured in @jcellsci.bsky.social, she plans a postdoc with piano lessons and exciting cell biology!

TFG forms a hollow condensate to shape the ER-Golgi interface, acting as a molecular filter - passing COPII anterograde carriers while excluding COPI retrograde coats. Hydrophobic patterning enables selective transport. Cool work from Andreas Ernst lab @UCSD! Nat Comms www.nature.com/articles/s41...

On April 19, 1898, Camillo Golgi described what we now know as the #Golgi apparatus - a structure so pretty and unusual it took over fifty years for the scientific community to agree it wasn’t just an artefact.
April 19 is #GolgiDay! Mark your calendars to honour the best organelle.

Interesting preprint from Junjie Hu on regulated Golgi retention: LEPROT and LEPROTL1 act as COPI adaptors that transiently retain transmembrane receptors in the Golgi - making a checkpoint for proper modification. Without them, proteins 'leak' out of the Golgi.

Amazing microscopy from Bewersdorf/Rothman groups shows golgins in a striking four-layered structure at the Golgi rim. Biochemical reconstitution shows long filaments. Does a Golgi stack form due to golgin filament properties rather than dedicated stacking proteins?

Interesting preprint from Boyce lab. During mitosis ER-Golgi traffic halts, Sec24C loses O-GlcNAc and disperses from ER exit sites. Phosphorylation also regulates Sec24C stability and localization, highlighting coordinated roles for phosphorylation and glycosylation.
