Here we argue that they have a lasting value, both in enabling discovery and in cultivating scientific thinking, by training researchers in systems reasoning, integrative thinking and independent inquiry.

Depends on what you want/need! We've removed a promoter with flanking guides and it worked well. Genotyping after in situ can be tricky though, if the deletion is bigger than 400bp. (DNA gets sheared)
