🙏🏻 for the highlight @frenchdcsociety.bsky.social one important finding was the very efficient #vacuolar pathway in MZ #macrophages challenging the current view that #proteasome dependent pathways (active in DC1) are prominent to generate antigenic peptides for #MHC-I through #cross-presentation

Splenic metallophilic marginal zone macrophages (MMMs) prime CD8+ T cell responses via MHC class I cross-presentation. Cross-priming by MMMs protects against a splenic tumor in the absence of dendritic cells.

Metallophilic marginal zone macrophages cross-prime CD8+ T cell-mediated protective immunity against blood-borne tumors
Metallophilic marginal zone macrophages are positioned strategically within the spleen to process internalized blood-borne materials. Mauvais et al. find that these CD169+ macrophages cross-prime CD8+ T cell responses to internalized antigens using a proteasome-independent pathway, contributing to antitumor immunity independently of type 1 conventional dendritic cells.

🚨 New: Spleen-resident macrophages (MMMs) can directly activate CD8+ T cells to fight tumors – independently of dendritic cells. They use a vacuolar MHC-I pathway & need Batf3 for this antitumor response.
