Soluble Notch agonist have the potential to bypass the need for feeder cell lines in the generation of therapeutic T cells and ILCs. Huge potential and can’t wait to try it out (if it’s available to researchers)!

The authors further showed that mice from JAX lab (where many of us get lab mice from) lack this commensal. Differences in mycobiome can dramatically influence immunity.

In @nature, Liao et al showed this commensal promotes type-2 immunity in the gut via IL-33/ST2, influencing protective anti-helminth responses and food allergy models.

A naturally occurring fungal symbiont that is missing in some lab mice maybe be contributing to inter-facility variation in type-2 immunity.

We used BD Cytofix/perm, Foxp3 fix/perm or 2% PFA fixation and these were all compatible with 2W1S tetramer staining and downstream cytokine analyses by flow

I can still see some after stimulation, so it’s not like the stimulation will completely negate tetramer staining (at least for 2W1S-MHCII tetramers). Worth a try!

Agree with the above and I will add that at least for class 2 tetramers, it will depend on the specific tetramers that you use. Unfortunately a lot of the class 2 tetramers are not great compared to class 1, and in my experience both the MFI and % positives may reduce after stimulation, but

The power of spatial information! ILC2s specifically in the middle of the colon express microbiota-induced genes that are important for the structural adaptation of this part of the gut. Loads of spatial data across different states (homeostasis vs inflammation) for the community.
