Ja, war normale SARS-CoV2-Impfung von BNT oder Moderna und da gibt es ja umfangreiche Sicherheitsdaten. Geht hier "nur" um zusätzlichen Effekt wenn besagte Tumor-Patienten neben der Immuntherapie mRNA-Impfung bekommen. Aber das könnte weitreichende Konsequenzen haben.

5/ Ergebnisse sollen ab Ende des Jahres in prospektiver randomisierter Phase 3-Studie überprüft werden. Wenn sie sich bestätigen könnte Effizienz durch Design noch geeigneterer mRNA-Vakzine/Therapeutika und gezielteres timing der Immunstimulation vermutlich weiter optimiert werden.

4/ Bias wie healthy vaccine effect ist durch Berücksichtigung zahlreicher confounder unwahrscheinlich. Hauptmechanismus scheint die mRNA-spezifische Aktivierung des Immunsystems in Kombination mit Ausschalten der Verteidigungsmechanismen der Tumoren über Checkpoint-Inhibitoren (ICI) zu sein.

3/ Das wurde jetzt an ca. 1000 Pat bestätigt. Hierbei hatten Pat. mit Bronchialkarzinom, die innerhalb von 100 Tagen vor Immuntherapie mit mRNA-SARS-CoV2 geimpft wurden etwa doppelt so hohe Überlebenszeiten wie Kontrollen, ähnliches fand sich bei Malignen Melanomen.

2/ Die Autoren hatten in früheren Studien zunächst in Tiermodellen festgestellt, dass mRNA-Impfstoffe zu Sensibilisierung sonst therapieresistenter Tumoren führt, die sie durch Immuntherapien behandelbar machen.

1/ Neue Studie aus den USA könnte Immuntherapie bösartiger Tumoren entscheidend transformieren. Studie ist retrospektiv aber scheint, soweit man bisher veröffentlichten Daten entnehmen kann, sehr solide und und basiert auf sorgfältigen präklinischen Vorstudien.

Only the UK had a good idea of its infections and yet I am not aware of a single study using it to determine IFR of covid by age

A study was published today by Ioannidis et al on lives saved by vaccines and their IFR is massively lower than the real data based on from the UK

Would still be a big success of Covid vaccination, but don't forget that the focus of the study by (guess who?) is to claim the the number of lives saved is much lower than other studies have calculated.

Actually Moderna reported higher VE against symptomatic disease as a result of their Phase 3 study (not published yet, woul be interesting to see results in detail). The higher NAb-titers by mNEXSPIKE had been shown in their Phase 2a study, published in January 25.

Sehr wichtige Studie der Gruppe von @deeptabhattacha.bsky.social, die zeigt, dass nicht Imprinting sondern geringe Immunogenität der Omikron- Epitope für schwächere Omikron-spezifische Immunantwort verantwortlich ist. Ergebnisse können enorme Bedeutung für zukünftige Vakzinentwicklung haben.

TL;DR, as SARS-CoV2 Spike epitopes drifted, many became poorly immunogenic, irrespective of antigenic imprinting/prior immunity.

I've been yelling for a while about how imprinting is not the problem with our immune responses to Omicron, and the issue is much more about the intrinsic immunogenicity of Omicron's spike protein (which is poor). This freshly published study provides pretty definitive proof of that 🧵

Eben "so rasch" ist relativ. Dürfte vor allem für gesellschaftliche Krankheitslast ziemlich irrelevant sein. Vermute dass Hauptfaktor für steigende Abwasserdaten waning immunity nach langer Zeit relativ niedriger Viruszirkulation ist.

Habe bisher nur Kapitel 7 "Das vernachlässigte Gain of Function-Labor der Natur" gelesen. Ist alleine das Geld wert, wenn man die (leider ) hochgradig relevante und massiv instrumentalisierte Diskussion über zoonotischen Ursprung oder Lab leak von SARS-CoV2 besser verstehen möchte.
