Using RT-qPCR as the reference standard on 197 nasopharyngeal swabs,
the Pluslife assay showed a positive percentage agreement (PPA) of 96.30% and a
negative percentage agreement (NPA) of 99.30% (κ=0.962, P<0.001). The EasyNAT
assay demonstrated a PPA of 96.30% and NPA of 98.60% (κ=0.949, P<0.001).

In Romania (2024), backyard birds—chickens, geese, ducks, pheasants—did not sustain circulation. An H5N1 outbreak ended quickly, with RT-qPCR turning negative. Rapid responses plus biosafety can stop local persistence and viral spread. 3/8

Nobody cares about trad qualifications in the biohacking world. 😋

But e.g. trad qualifications: B.S., ~3 yrs of wet lab and surgical experience in academia including gonadectomies, brain harvesting, stereotactic brain surgery, brain staining, RNA extraction (RT-PCR), qPCR, etc.

had tissue-level analysis done to examine which cells took up the mRNA-LNPs. If you guessed it was predominantly antigen-presenting cells that took them up (as has been shown in every study looking at this question), you would be right. Interestingly, some uptake was also seen in B cells.

Little scientist ego boost today when my custom-designed qPCR primer pairs for GAPDH were all within acceptable efficiency range, except for the IDT "ready-made" pair that I purchased on the side, which was by far the least efficient. Also - despair that people are wasting money on a bad pair!

In both mouse & human data sets, B:H ratios aligned with qPCR data and diet-derived read normalization.

Additionally, we found data from a milk microbiome study, where they used a spike-in to estimate total bacterial biomass. Spike-in and B:H biomass estimates showed almost perfect agreement

Other vials were opened and used, with some remenants refrozen and shipped. This will affect the quality of the mRNA.

Plasmid DNA was detected via qPCR with Ori, Spike and SV40 promoter probes. Strangely, the 3 probes give different content of DNA, which should not happen.
