Fungal Friends - Some of you may have seen the recent preprint on chromosomes in Neurospora nuclei. I have something to say about that, but waiting while bioRxiv moderates the comment

Fay Ajzenberg Selove - if you have done any work with light nuclei, you've cited her papers. So fortunate that she was part of my academic path, especially when I was dealing with mental health issues as an undergrad.

Can you move to Cut&Tag? We also had issues with Run variation. In the end, replicate variation for either assay was normalized by spiking in 10% mouse nuclei into human assays and 10% human nuclei for mouse assays.

It blew me away when I first learned this too. The next amazing thing is that transcription factor concentration gradients cause each region of nuclei to specialize into different cells, thereby dividing the insect into segments. I'm still trying to understand how gradients accomplish this.

A fertilized insect egg doesn't repeatedly divide like a vertebrate egg. Instead, its nucleus duplicates itself thousands of times (6000 in fruit flies). These nuclei migrate to the cell periphery, where the membrane folds around them to form cells. (Alberts et al., Molec Bio of the Cell, 7th ed)

1. Endogenous CRISPR knock-in tag
2. Promotor transgene fused to Histone
3. Endogenous CRISPR knock-in dual color tag with self-cleaving peptide T2A fused to Histone.
