Paneth-like transition drives resistance to dual targeting of KRAS and EGFR in colorectal cancer
Zhang et al. show that CRC cells adopt a Paneth-like state to evade dual KRAS and EGFR inhibition. They identify a SMAD1-FGFR3 axis that drives lineage plasticity and MAPK reactivation, conferring resistance. FGFR3 co-targeting blocks this transition, offering a strategy to improve the durability of KRAS-EGFR combination therapies.
