🔍 As expected, the strongest trans-regulation of TCRs occurs in the MHC region.
What’s interesting 🤨 is that HLA-associated V genes were less stable across treatment. Notably, patients with HLA-regulated TCR clones showed improved overall survival.

On the pathogen side, we observed that the interacting strains have altered redox states due to thioredoxin reductase mutations.

Taking this observation further, we experimentally assessed the responses of human macrophages from TB donors with different host genotypes to different Mtb infections. We observed that hosts with interacting variants exhibited stronger interferon gene signatures.

Strikingly, we identified a unique subclade of Mtb lineage 2 that interacts with human variants within the MHC region.
