Now, there are over 100 types of sarcoma (!), it is an incredibly diverse family of tumors. They are all rare and ultra-rare cancers, many without any type of model that allows us to study the biology and develop effective therapies.

Over the past few years, we have developed an approach to rapidly establish and screen 3D tumor organoids. We have deployed this approach to study fast growing or indolent malignant tumors. Now we translate it to cutaneous neurofibromas.

There is no systemic therapy, and if the tumors cause any issue they need to be removed surgically for the most part. It is also a very challenging tumor type to model in the lab, as it is formed by many different cell types and abundant extracellular proteins.

Neurofibromatosis type 1 is a common inherited disorder caused by alterations in the NF1 gene. One of the most penetrant features of the disease is the formation of these benign tumors, cutaneous neurofibromas, in the dermis. They can be anywhere in the body, and numerous.

Here’s the thing about benign tumors. While they may not metastasize, they can still cause substantial health issues, particularly when they number in the hundreds, or thousands such as the case of cNFs in Neurofibromatosis type 1 patients.
