Using a variety of approaches, including fluorescence microscopy, electron microscopy & tomography, we found that Salmonella were in damaged SCVs & hyperreplicating in the cytosol of human macrophages lacking inflammasome responses! Salmonella also replicated more within SCVs. (8/n)

Salmonella normally replicates in vacuoles. In human macrophages lacking inflammasomes, we saw 'jackpot' cells with hyperreplicating Salmonella, reminiscent of beautiful findings by @leighknodler.bsky.social & others that Salmonella can escape vacuoles to hyperreplicate in the cytosol of IECs. Excited about the 33rd Annual Philadelphia Infection & Immunity Forum today, organized by our local Eastern PA @asm.org student branch! We have a record 149 registered attendees, 62 poster presentations, 6 trainee talks, & 3 keynote speakers (Boris Striepen, Jorge Galan, & Ellen Foxman)!
