By integrating this to the MoClo standard, we enabled standardized, automated assembly and rapid testing of genetic combinations - resulting in more than 30 new synthetic genetic elements. We demonstrated a proof of concept by introducing a synthetic photorespiration pathway into the chloroplast!

We developed an automation workflow to generate, handle, and analyze thousands of transplastomic strains in parallel. We expanded the available selection markers and reporter genes and systematically characterized over 140 regulatory parts.

In our new study, we established Chlamydomonas reinhardtii as a prototyping chassis for chloroplast synthetic biology. This fast-growing alga has a single chloroplast per cell and can be cultivated in high-throughput formats, making it an ideal testbed.
