No, this was me messing around independently. Only deskew; no rotation. It is actually surprising how little difference it makes.

Dilute more than recommended. At least 1:4000. Probably even more for the SPY-DNA dyes.

... and a projection perpendicular to the light sheet, so not truly a view from the top but at 38 (or 52) degrees ... this is still giving me a headache.

well ... maybe, if someone invites me 🤪. I am realizing that one full scan of three channels of a relatively flat volume is 1GB ... this is going to present some challenges for time-lapse.

Yep. That’s the question. Tempted to test with nanocages because it it is really this bright and stable would be very useful… however, @erin-rod.bsky.social, is this thing a dimer?
