I lead a team studying musculoskeletal physiology, development, and healing. We study how the skeleton grows and how connective tissues guide this process.

Grateful to attend the Myogenesis GRC in Tuscany this week, where I met loads of new people, learned a ton about MuSCs and fibroblasts, and gave a short talk on our optogenetics work

We had three talks and three posters including late breaking talk by Nicole, Stephs first podium at ORS, and Allies poster in the Undergraduate session! + @sssteltz.bsky.social competed in the Tendon Section 3min thesis!
Our chair, Dr. Caird, also received the Womens Leadership Forum Award!!

We were relieved to find that, while the ChR2-EYFP muscle was functional impaired compared to wildtype controls, the ChR2-only muscle was physiologically normal, and also reached peak twitch force more quickly than the ChR2-EYFP muscle when using blue light to initiate contraction.

She also characterized muscle structure, using histology and TEM, and found significantly smaller fibers in the ChR2-EYFP muscle compared to Cre-negative and ChR2-only mice. Syeda also did not find vacuoles in the ChR2-only muscle, which was a relief.

Syeda set off to identify if and how the ChR2-EYFP strain differed, functionally, from this new ChR2-only strain that @brianglancylab.bsky.social developed. She characterized muscle function using a battery of physiological tests by measuring muscle force in response to electrical stimulation

He shared with us some confocal and transmission electron microscopy images that showed aggregation and vacuoles in the sarcolemma of myofibers in the Ai32 mice when using a different muscle-related Cre line than ours, suggesting an accumulation of protein on the membrane 😬 We found the same thing!

Following a tweetorial on our CTR pub in 2020, @brianglancylab.bsky.social reached out via DM to chat about these mice. What we thought was general eYFP expression in muscle (shown in green in C) was something his lab had been interested in as well- and they found some problematic issues w/ it.
