Session 11: Last but not least Raquel Lieberman - Rare coding mutations in MYPC OLF cause glaucoma. ECM ring where the sclera begins in your eye. Mutant myocilin accelerates trabecular meshwork dysfunction. Toxic GOF missense mutation in OLF domain. Aggregates in ER. #LorneProteins2025
Not all rare mutations cause glaucoma, some are neutral polymorphism. Nomad database of variants and clinical significance. Using physicochemical intuition is insufficient. GenAI 50% correct. Mutation/misfolding relationship underpins better clinical outcomes for kids. Genotyping is not standard of care. How many and which point variants are pathogenic? Biophysical signature of a disease variant? structure/stability/dynamics/folding. Incompatible residue change? Take 1 in 10000 from Nomad, in vitro, xtallisation - look like wt. Age at diagnosis vs thermal stability. Avg thermal stability pathogenic 42 deg C. Cut off 47 deg? One example fell out of the clusters. A427T 47.3 73 yo; carried by asymptomatic and symptomatic family members. Amyloid-like aggregates. Similar to wt structure. Other factors? Environmental... Solution NMR. res assignment, OLF is not at equilibrium. Stabilises in 90 days. D380A mild variant mt 46.6, I499F Mt 42.8. OLF mutants are destabilised in the core. Midpoint of unfolding urea induces inside out unfolding. Mutants chemical shift perturbation mimics midpoint unfolding NMR spectra, so mutation causing partial unfolding of half of the fold proximal to the short alpha helix. Do dusease variants affect the unfolding pathway? Complex unfolding landscape. Loss of internal Calcium is an early unfolding event Inherited coding mutations often cause glaucoma. OLF unfolds from the inside out. External strands still intact. Partial folded to fibrillar sink. Tribecular meshwork cells can't handle this.

TIM50 core of TIM23 complex in the inner membrane. N term of PINK1 traps tom40 and tim23 in a supercomplex between membranes across the intermembrane space. Trapped supercomplex and express pink1-flag. Mito isolation, detergent exchange, sec and cryoEM. HSPs and Vdacs in the complex too. 6 pore complexes in all particles. 3.1 Å Pink1. N term inside the barrels of tom 40. Vdac 2 dimer is centre and 2 tom 40 complexes. First structure of human VDAC2! TOM5 connecting VDAC2 to Tom40.
