2/2 and how high-throughput you need the chemistry and assay to be. It’s anything but trivial for chemistry that has a lot of components and for whole cell assays. If your chemistry doesn’t work in DMSO, or compounds are unstable or for any other reasons it can be not as easy as it sounds!

1/2 IME, there’s actually a fairly large resource lift before “just putting compounds on cells” to validate DtB assays. Of course, ppl sometimes just go ahead and try DtB, but that usually tells you very little. Readouts always depend on how sensitive the assay is, which controls are the right ones,

How about paraffins? I assume that if your reaction is soluble in Ph2O, those could be good enough…
