Topological barriers ahead of the replisome lead to bursts of replisome rotation. To our surprise, excessive rotation does not reduce replisome speed, but does have negative consequences for coordination by increasing pausing, reducing processivity, and altering polymerase exchange dynamics.

For the first time, we were able to spatially resolve the parental, leading and lagging strands in real-time during DNA replication using fluorescence microscopy.
