Here’s an example showing how you can reproduce a classic Scheres-type RAM stem-cell ablation experiment using Bax.

✨ To make this possible, we developed LRTracker (stable) and iLRTracker (inducible) with versatile reporters—powerful tools to study how e.g. environment controls LRP formation and the decision between LR and cambium fate. 3/3

During times like this, it is good to read or listen to a book. Below is my strong recommendation. What do you recommend?

Just type you gene name, and you'll see how your gene is expressed in secondary tissue, and how it corresponds to our uMAP. Here is an example of a nice vascular cambium marker we identified. 6/x

Hiroyuki discovered that phloem parenchyma gradually obtains periderm identity, and this transformation can be accelerated by jasmonate treatment or wounding. Thus, it appears that a key function for the 'fluffy' phloem parenchyma cells is to be a reservoir for the protective periderm. 4/x

Combining the scRNAseq data, reporter analysis and lineage tracing Hiroyuki and Munan identified hierarchical cell fate determination map of all the cell types in the secondary tissue. 3/x

To obtain the detailed map, Munan and Meeri validated the scRNAseq data with close to 100 fluorescent reporter lines. Parenchyma tissue in phloem and xylem is more diverse than we previously though, and more dynamic… 2/x

In our Tansley review, we attempted to establish the stem cell concept in vascular cambium the way we see it. I hope that this is useful also for the other cambium enthusiasts. nph.onlinelibrary.wiley.com/doi/10.1111/...

Xin made two version of the LR tracing tool: inducible iRootTracker and constitutive RootTracker. You can monitor LR formation through agar plate under stereo fluorescent microscope and screen several roots/min, and the tracker appears to be very sensitive (nM scale IAA/BAP) 4/x

Overall, the fate of XPP and LRP cells is very plastic, determined by the balance between auxin and cytokinin. 3/x

New preprint from our lab. Xin Wang developed a handy genetic tool called #RootTracker to follow fate of lateral root primordia (LRP). He showed that arrested LRPs gradually obtain cambium identity and contribute to the secondary growth. www.biorxiv.org/content/10.1...
