It doesn't need to be something complicated or revolutionary. Just something that feels authentic and that you believe, so it can be better conveyed to the audience you aim to reach.
Another great example would be #Berserk, which asks if you "Can you find meaning in a world devoid of hope?" This way, the story can work its way towards providing an answer to that thematic question. Show all forms of sides and arguments, but then provide an answer YOU believe.
This way you don't pick something vague like "Justice" or "Love" and get something tangible the audience can understand. For example, in Arcane, it would be "Can the cycles of trauma and violence be broken, or are we bound by the mistakes of the past?"

Vander and Silco - Led said coup, traumatized for life
Ekko - Loses all friends and father figure
Caitlyn - Goes to investigate Shimmer, loses mother
Viktor - Being born in Zaun might be why he sick
Jayce - Makes the divide worse by accident
Think about how the tensions between Piltover (High Society) and Zaun (Lower Class) are the reason for every single problem, at least in S1 (I love you S2, but after S1 10/10, you dropped into a confortable 7/10). Every character has their lives shaped by this looming thing, no matter who.
I think a lot of beginner writers forget to give memorable failures to their characters, to constrast with their victories.
She's a strong fighter, accomplishes a lot in her time, but the core moments that define her and make her memorable are how she fails to stop a gangster from getting her sister and failing to save said siter from turning into a monster.
I think that the failures of a character make them more interesting than their victories. Consider Vi, from League of Legends.
