While progress has been made in understanding the etiology of Major Depressive Disorder (MDD), significant challenges persist, including limited representation of ancestral diversity in study cohorts and substantial heterogeneity among phenotypes. The Latin American Genomics Consortium MDD Working Group (LAGC-MDD) is actively addressing these limitations. In this presentation, we will provide an overview of depression assessments, harmonization efforts, and genomic analysis pipelines. Datasets have been aggregated to perform the first large-scale genome-wide association study (GWAS) meta-analysis of depression phenotypes in individuals of admixed Latin American descent. An initial qualitative analysis was performed to assess MDD phenotype heterogeneity. As of May 2024, data have been aggregated from 33 datasets spanning 6 countries (Brazil, Colombia, Mexico, Peru, United Kingdom, United States). Our initial examination (meta-analysis n=135,000 cases; 240,000 controls) revealed that depression phenotypes were assessed using various measures, ranging from single-item self-reports to clinical interviews. Medical records (ICD codes) were the most prevalent type, followed by the CESD self-report scale. Large datasets are essential for conducting well-powered analyses, yet they may introduce heterogeneity. Our planned analyses aim to assess and model heterogeneity across depression phenotypes using psychometric and genomic approaches to parse this variability and maximize analytical power. The LAGC is actively engaged in enhancing data collection and capacity building within Latin American communities. Given that MDD ranks among the most prevalent mental health disorders in Latin America, it is crucial to mobilize large-scale, targeted consortia efforts to advance our understanding of this debilitating disorder within these under-studied populations.

We had an early Brain Awareness Week this year. We visited an elementary school where we asked them to invent a brain for an animal with any super power they wanted. The resulting brains and explanations were extraordinary!!
