Finally out in print! Long time project with @olexandr.bsky.social and the PHENIX group. Refinement of X-ray and CryoEM structures, using Machine Learned Potentials as the back end to help fitting. This fulfills the old dream of doing QM refinement, but very very cheap www.nature.com/articles/s41...

FYI I tried it on a larger membrane protein for which the 2D classes were not high res (not enough particles) but I hoped for a miracle. The miracle didn't come, I ended with a high res bad model, a negative control I guess.

Initial attempt at replicating in relion (parameters in next post). This is for Aca2-RNA, using a 100k subset of the 2D-classified particles (no prior 3D cleanup).
A 1-class ab initio in relion, then local refinement in relion (1.8deg searches+blush) gives a nominally 3.3Å map; 3.5 Å w/out blush.

Soon... a new building for our laboratory, with 1400m² extra and renovated space to host new equipements such as cryoEM, CryoFIB-SEM, optical microscopies in biosafe confined environment, and a synthetic biology labs.

Using Cryo-EM❄️ on an amazing sample, we found that YidC is recruited to the back of SecYEG during late-stage substrate insertion —supporting that the "back-of-Sec" route observed in eukaryotes also exists in bacteria 🦠.
