Interestingly, we have collaboration with #Barntumörbanken where we collect these paired primary and relapse samples to study the tumor evolution and also focus on understanding best treatment strategies for these patient #ISRVT25
Btw, I am a second year PhD student working on DMG. We are mostly using patient biopsies and trying to understand different treatment options for these patients #ISRVT25
They are mostly occurring in pre-school children growing in the thalamus, pons or the brain stem. #ISRVT25
Diffuse Midline Glioma is one of the most aggressive childhood brain tumors, with very limited treatment options.
They are known to have signature mutations in histone 3 variants. Additional mutations also include PDGFRA, TP53, ACVR1 and PI3K mutations #ISRVT25
And additionally is also quite nice because this was one can also setup high throughput drug screening with these cultures. They are more transactional than 2D cultures since they have a microenvironment.
This is the reason why tumoroid or organoid cultures have become so attractive in these kind of studies in the last few years
One of the reasons why it is important to have the 3D microenvironment around these cells to get a better overview on how they are interacting or establishing themselves or even surviving (3/5)
Most of these tumor cultures tend to die or loose their properties when cultured in 2D.
One of the most common examples being Sonic-Hedgehog Medulloblastoma
